Axonal transport is a highly complex process which is regulated by a family of small GTP-binding proteins. GTP-bound members of the Rab and Arf families act as molecular switches, regulating the activation of the motor proteins. Rab proteins mediate membrane trafficking events at the axon terminal, while Arfs promote the priming of cargo vesicles for transport towards the growing tip. The most well-characterized member of the Rab family is Rab3, which is involved in the transport of synaptic vesicles at the developing and regenerating neuromuscular junctions. Rab3 is localized to the growth cone, where it regulates the formation of new vesicles, and its activity is controlled by several regulatory factors including GTP-ase activating proteins, guanine nucleotide exchange factors, nucleotide-binding proteins and guanine nucleotide dissociation inhibitors. A recent study demonstrated the involvement of the small GTPase Arf6 in axonal trafficking events in vivo. In this study, the axonal localization and mode of action of Arf6 is described, as well as its biological role in neurons.The proposed work deals with the control of pulmonary surfactant production and secretion.
